Target
Sodium-dependent, high-affinity amino acid transporter that mediates the uptake of L-glutamate and also L-aspartate and D-aspartate (PubMed:7914198, PubMed:7521911, PubMed:8857541, PubMed:26690923, PubMed:21123949) . Can also transport L-cysteine (PubMed:21123949) . Functions as a symporter that transports one amino acid molecule together with two or three Na (+) ions and one proton, in parallel with the counter-transport of one K (+) ion (PubMed:7521911, PubMed:8857541, PubMed:26690923) . Mediates Cl (-) flux that is not coupled to amino acid transport; this avoids the accumulation of negative charges due to aspartate and Na (+) symport (PubMed:8857541, PubMed:26690923) . Assay Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Excitatory Amino Acid Transporter 3 (EAAT3) . Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Excitatory Amino Acid Transporter 3 (EAAT3) . Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Excitatory Amino Acid Transporter 3 (EAAT3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Excitatory Amino Acid Transporter 3 (EAAT3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
