D61 is a potent STING agonist with EC50 values of 52.9 nM and 116 nM for inducing IFN-β and SEAP reporter activity in vitro. It activates the TBK1-IRF3 pathway in monocytes and demonstrates in vivo antitumor efficacy in a CT26 colon cancer model at 0.25 mg/kg without weight loss.
