Mt KARI-IN-2 (5b) is a potent and selective inhibitor of Mycobacterium tuberculosis ketol-acid reductoisomerase (Ki = 2.02 μM) with antitubercular activity (MIC = 0.78 μM against Mtb H37Rv) and low cytotoxicity. This compound is a valuable tool for investigating Mtb KARI's role in branched-chain amino acid biosynthesis and for in vitro antimycobacterial mechanism studies.
