WAY-265497 is a potent ATP-binding site-targeting inhibitor of Cdk4/D1 kinase activity with an IC₅₀ of 30 nM. It inhibits HCT116 and MCF-7 cancer cells with IC₅₀ values of 0.73 and 0.88 µM, respectively. It inhibits Cdk2/E and Cdk1/B at higher concentrations with IC₅₀ values of 1.6 µM and 25.3 µM, respectively.
