Background
DiR (DiIC18 (7) ) is a lipophilic, near-infrared fluorescent cyanine dye with the molecular formula of C63H101IN2, molecular weight of 1013.4, and CAS number of 100068-60-8. This dye is often used to label cytoplasmic membranes. The two 18-carbon chains of DiR are inserted into the cell membrane, resulting in specific, stable cellular staining with little cell-to-cell dye transfer. The combination of DiR (near infrared fluorescence) and other cell membrane fluorescent dyes such as DiI (orange fluorescence), DiO (green fluorescence), and DiD (red fluorescence) provides an effective tool for multicolor imaging and flow cytometry analysis. After DiR staining, fixation with paraformaldehyde (no other reagents such as methanol) can be performed, but the process of permeabilization after staining is not recommended." In addition, plasma membrane staining was also well performed after fixed permeabilization with 0.1% TritonX-100 at room temperature. Using 100 μL of staining working solution at a concentration of 5 μM, 5 mgof working solution could be used about 9,867 times
