Caspase family of cysteine proteases has been shown to play a key role in apoptosis. Mammalian caspases can be subdivided into three functional groups: initiator caspases (Caspase 2, 8, 9 and 10), executioner caspases (Caspase-3, 6 and 7), and inflammatory caspases (Caspase 1, 4, 5, 11 and 12) . Caspase-4 (also known as Ich-4, ICErelII, TX) is a member of the caspase-family of cysteine proteases. The active caspase-4 is a heterotetramer consisting of two large and two small subunits.

Caspase 4 Assay Kit (Colorimetric) is based on the hydrolysis of the peptide substrate Ac-LEVD-pNA (acetyl-Leu-Glu-Val-Asp p-nitroanilide) by caspase 4, resulting in the release of the p-nitroaniline (pNA), which can be quantified using a spectrophotometer or a microtiter plate reader at at 405 nm.
