Caspase-1 Assay Kit (Colorimetric)
Caspase-1 Assay Kit (Colorimetric) provides a simple and convenient method for detecting the activity of caspase-1.
Product Specifications
Background
Caspase family of cysteine proteases has been shown to play a key role in apoptosis. Mammalian caspases can be subdivided into three functional groups: initiator caspases (Caspase 2, 8, 9 and 10), executioner caspases (Caspase-3, 6 and 7), and inflammatory caspases (Caspase 1, 4, 5, 11 and 12) . Caspase-1 (ICE, interleukin-1β-converting enzyme) is the mammalian caspase responsible for the proteolytic conversion of the proforms of two inflammatory cytokines, interleukin-1β and IL-18 into active cytokines, initiating a proinflammatory response. It also cleaves Gasdermin D into its active form, which leads to pyroptosis.

Applications Notes
Caspase 1 Assay Kit (Colorimetric) is based on the hydrolysis of the peptide substrate Ac-YVAD-pNA (acetyl-Tyr-Val-Ala-Asp p-nitroanilide) by caspase 1, resulting in the release of the p-nitroaniline (pNA), which can be quantified using a spectrophotometer or a microtiter plate reader at at 405 nm.
