EliKine™ Human IL-1β ELISA Kit

Interleukin 1β encoded by IL-1β gene is a member of the interleukin 1 cytokine family. Interleukin 1β is produced by activated macrophages as a proprotein, which is proteolytically processed to its active form by caspase 1 (CASP1/ICE) . Interleukin 1β is an important mediator of the inflammatory response, and is involved in a variety of cellular activities, including cell proliferation, differentiation, and apoptosis. The induction of cyclooxygenase-2 (PTGS2/COX2) by Interleukin 1β in the central nervous system (CNS) is found to contribute to inflammatory pain hypersensitivity. IL-1β gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2.

EliKine™ Human IL-1β ELISA Kit has high sensitivity and excellent specificity for detection of Human IL-1β. No significant cross-reactivity or interference between Human IL-1β and analogues was observed.

EliKine™ Human IL-1β ELISA Kit employs a two-site sandwich ELISA to quantitate IL-1β in samples. An antibody specific for IL-1β has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IL-1β present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL-1β is added to the wells. After washing, proprietary EliKine™ Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL-1β bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Do not mix components from different kit lots or use reagents beyond the kit expiration date.• Allow all reagents to warm to room temperature for at least 30 minutes before opening.• Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.• It is recommended that all samples and standards be assayed in duplicate or triplicate.
