TAOK3, a kinase, regulates the p38/MAPK14 and MAPK8/JNK cascades. It activates the p38/MAPK14 cascade, particularly in DNA damage-induced G2/M transition, likely through MAP2K3 and MAP2K6 phosphorylation. TAOK3 inhibits basal MAPK8/JNK activity and dampens its response to epidermal growth factor.
