PRTN3 (Human) ELISA Kit
Proteinase 3 (PR3, PRTN3) is a serine protease that belongs to the trypsin family of serine proteases. It is primarily stored in the azurophilic granules of neutrophils. However, it is also present in secretory vesicle membranes, monocytes, endothelial cells, epithelial cells, and some human cell lines (HL60 and SK-RC-11). One of the main physiological functions of PRTN3 is the proteolytic degradation of extracellular proteins at the site of inflammation. In addition to extracellular proteins, it also degrades proteins such as elastin, hemoglobin laminin, collagen type IV, and fibronectin. Besides proteolytic degradation, PRTN3 also modulates the activity of platelets, endothelial cells, dendritic cells, and inflammatory mediators (C1 inhibitor, TNF-α, TGF-β, IL-2, IL-8). PRTN3 is the main target of anti-neutrophil cytoplasmic antibody (ANCA), a type of antibody associated with systemic vasculitides, myeloid leukemia, and Wegener’s granulomatosis. Binding of PRTN3 with ANCA indicates the extent of cellular and tissue damage and development of the diseases. Hence, PRTN3 could be considered a promising therapeutic target for inflammatory diseases. BioVision’s PRTN3 (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of PRTN3 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of PRTN3 in the sample or standard.
