Magic™ Cell-free Protein Expression in E. coli Lysate

Magic™ Cell-free Protein Expression in E. coli Lysate Kit can be used for in vitro recombinant protein synthesis in high levels. It is noticed that this kit can synthesize proteins from both circular plasmid and linear templates (e.g. PCR products). In terms of the template, it should contain a T7 promoter. The initial protein synthesis reaction is composed of E.coli extract, reaction buffer and amino acids. After initiating the protein synthesis reaction, supplemental buffer is added in the reaction to replenish the necessary components which are depleted or degraded during protein synthesis. Supplement with the buffer allows continuous cell-free protein synthesis, resulting in recombinant protein production in a higher yield in up to 3-6 hours. We recommend wearing gloves and using nuclease-free tubes and tips to avoid introducing nucleases to your samples.
