Background
CD62L is the equivalent of the mouse MEL-14 with a molecular weight of 80 kDa. CD62L is on 60-75% of peripheral blood lymphocytes, Approximately 70-80% of T cells, 10% of thymocytes and most B cells. CD62L reacts with monocytes, granulocytes and a subset of NK cells. The selectins represent a family of adhesion molecules, expressed by leukocytes and endothelial cells that are involved in the regulation of leukocyte traffic. Human leukocyte L selectin is also called LAM-1, LECAM-1, or gp90MEL-14. This antibody to L selectin blocks greater than 95% of lymphocyte adhesion to peripheral lymph node high endothelial venules.

Assay Principle
PBMC: Add10 µL of MAB/10^6 PBMC in 100 µL PBS. Mix gently and incubate for 15 minutes at 2º to 8º C. Wash twice with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. WHOLE BLOOD: Add 10 µL of MAB/100 µL of whole blood. Mix gently and incubate for 15 minutes at room temperature (20ºC). Lyse the whole blood. Wash once with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. See instrument manufacturer’s instructions for Lysed Whole Blood and Immunofluorescence analysis with a flow cytometer or microscope.
