Serum Amyloid A (SAA) is an acute-phase plasma protein that belongs to the family of apolipoproteins. Pro-inflammatory cytokines such as IL-6, TNF, TGF-β, and interferon-γ stimulate the production of SAA. The concentration of SAA increases 1000 fold within 5-6 hours of inflammation or injury. During the acute phase of inflammation, it forms a complex with plasma high-density lipoproteins, reroutes HDL transport to promote clearance of lipids from the injury site, and recycle lipids for tissue repair. The liver is the primary source of circulating SAA; however, SAA may also be produced by extrahepatic tissues. High levels of SAA have been reported in pathological diseases such as cancer, diabetes, obesity, Alzheimer’s disease, atherosclerosis, and rheumatoid arthritis. Increased levels of SAA also help predict the progression of COVID-19 disease. Therefore, SAA serves as an efficient biomarker for the treatment of human diseases. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of SAA in the sample or standard
