Fill the square culture vessels with the required amount of media and cover them with their caps. Ensure that the vent is clear and unobstructed to allow for proper airflow. Autoclave the square culture vessels for 20 minutes at 15 psi and 121°C. Let the vessels cool down, and the media solidify. Avoid direct contact with the vent during handling to prevent contamination. Transfer your plants to the media under a laminar hood. Regularly check the vent to ensure it remains clean and free of blockages throughout the culture period.
