Neuroregulins (NRG) are polypeptide growth factors, that bind to HER proteins, withNRG1 being the most studied of the four known proteins in this family. NRG1specifically binds to HER3 and HER4. NRG1 binding induces dimerization of HER3 withHER2, transphosphorylation, and activation of several pathways such as the PI3K (phosphoinositide 3-kinase) /AKT signaling pathway. Due to HER3 cell surfaceexpression in a variety of cancers and correlation with decreased survival, HER3 is aprospective therapeutic target for antibody-drug conjugate (ADC) development. It hasbeen suggested that elevated levels of NRG1 in the TME (tumor microenvironment) canbe considered as a predictive biomarker for the effectiveness of anti-HER3 antibodytreatments in cancer. Binding of NRG1 to HER4 play an important role in corticaldevelopment, and cardiomyocyte proliferation and regeneration, by inducingdimerization to HER2 and activation of multiple signaling pathways.

Perform a quick spin of the vial followed by reconstitution in distilled water to a concentration of not less than 0.1 mg/ml. This solution can then be diluted into other buffers.

The lyophilized protein is stable for at least 2 years from date of receipt at -20° C. Uponreconstitution, this cytokine can be stored in working aliquots at 2° -8° C for 3 weeks, or at -20° C for one year, with a carrier protein at 0.1% without detectable loss ofactivity. Avoid repeated freeze/thaw cycles.
