Background
HDAC6, or histone deacetylate 6, is a Class II member of the histone deacetylase family which is involved in lysine deacetylation. Lysine acetylation/deacetylation is a dynamic process involved in the regulation of a variety of cellular functions, similarly to phosphorylation/dephosphorylation. In addition to its function in histones, HDAC6 can also target HSP90 (heat shock protein 90), α-tubulin and cortactin. Its interaction with α-tubulin and cortactin allows regulation of microtubule and actin-dependent cell motility. Critically, it also regulates proteins involved in cancer immunotherapy, such as PD-1 (program death receptor-1) and PD-L1 (program death receptor ligand-1), and it thus involved in cancer progression and metastasis. HDAC6 has also been implicated in Alzheimer’s disease. Ricolinostat, an HDAC6 inhibitor, showed little activity when used alone, but showed clinical potential when combined with lenalidomide and dexamethasone in the treatment of refractory multiple myeloma. The development of new inhibitors specifically targeting HDAC6, in a disease-specific context, and a better understanding of its modes of action may open newer avenues of HDAC6-linked diseases.
