The VSIG-3:VISTA [Biotinylated] Inhibitor Screening Assay Kit is a chemiluminescent ELISA designed to measure the binding between VSIG-3 (V-set and Ig domain-containing protein 3) and VISTA (V-domain immunoglobulin suppressor of T-cell activation, also known as B7-H5) protein for screening and profiling applications. This kit comes with enough purified VSIG-3 (amino acids 22-244) and biotinylated VISTA (amino acids 33-194), streptavidin-HRP, assay buffer, and detection reagent for 100 enzyme reactions.Figure 1. Next, unbound biotinylated VISTA is washed away, and the plate is incubated with streptavidin-HRP. After a final wash, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader. Background

V-domain Ig Suppressor of T cell activation (VISTA, or B7-H5) is a type I transmembrane protein that functions as an immune checkpoint. V-Set and Immunoglobulin domain containing 3 (VSIG-3, or IGSF11) is a ligand for VISTA which, when bound on activated T cells, inhibits their proliferation and the production of cytokines and chemokines. VSIG-3 expression is elevated in many cancers, especially glioma and melanoma, so the blockade of the VSIG-3/VISTA pathway represents a promising potential cancer immunotherapy strategy.
