The CD47:SIRP-α[Biotinylated] Inhibitor Screening Chemiluminescence Assay Kit is an ELISA-based assay designed to measure the binding between CD47 and SIRP-α (Signal-regulatory protein alpha) protein for screening and profiling applications. This kit comes with enough purified CD47 (amino acids 19-139) and biotinylated SIRP-α (amino acids 27-370), streptavidin-HRP, assay buffer, and detection reagent for 100 enzyme reactions.Figure 1. After coating and blocking, biotinylated SIRP-α is added in an optimized assay buffer. Next, unbound biotinylated SIRP-α is washed away, and the plate is incubated with streptavidin-HRP. After a final wash, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader. Background

Signal-regulatory protein alpha (SIRP-α), also known as CD172a, is a cell surface protein expressed mainly by myeloid cells such as macrophages. Its receptor, CD47, is ubiquitously expressed on the surface of all cells and has been found to be overexpressed in some cancers. CD47 is a macrophage immune checkpoint, and binding to SIRP-α delivers a “Do Not Eat Me” signal to macrophages, leading to inhibition of phagocytosis of tumor cells. In the context of a tumor, this signaling aids in tumor evasion of the immune system. The levels of CD47 mRNA have been correlated to survival rate in solid tumors. Inhibitors of this interaction hold great promise for cancer immunotherapy. For instance, anti-CD47 antibodies have been shown to induce a phenotype switch from M2 to M1 TAM (tumor infiltrating macrophages) and target tumor cells. The use of anti-CD47 in combination with other therapies, such as chemotherapy, seem also to result in better outcomes.
