The PD-1[Biotinylated]:PD-L2 Inhibitor Screening Assay Kit is an ELISA-based assay designed to measure the binding between PD-1 (programmed cell death 1) and PD-L2 (programmed death-ligand 2) protein for screening and profiling applications. This kit comes with enough purified biotinylated PD-1 (amino acids 25-167) and PD-L2 (amino acids 20-219), streptavidin-HRP, assay buffer, and detection reagent for 100 enzyme reactions.Figure 1. Next, unbound biotinylated PD-1 is washed away, and the plate is incubated with streptavidin-HRP. After a final wash, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader. Background

PD-1 (Programmed Death Protein 1, CD279) is a transmembrane protein expressed on the surface of immune cells that binds to PD-L1 (programmed death-ligand 1) or PD-L2, inducing apoptosis of T cells and inhibiting immune responses. Cancer cells often overexpress PD-L1 or PD-L2, allowing them to suppress T cell activation, evade the host immune response, and proliferate. Thus, disrupting the PD-1 co-inhibitory pathways may be an effective approach to developing anti-cancer therapies, as well as other immune diseases like multiple sclerosis, arthritis, lupus, and type I diabetes.
