Autoclave the square culture vessels for 20 minutes at 15 psi and 121°C. Let the vessels cool down, and the media solidify. Avoid direct contact with the vent during handling to prevent contamination. Transfer your plants to the media under a laminar hood. Regularly check the vent to ensure it remains clean and free of blockages throughout the culture period.
