Background

MDM2 (Mouse Double Minute 2 homolog) is an E3 ubiquitin ligase that negatively regulates the tumor suppressor p53, controlling its stability and transcriptional activity. In the nervous system, MDM2-p53 signaling is crucial for balancing neuronal survival and apoptosis. Overactivation of p53, due to MDM2 dysfunction, can lead to excessive neuronal loss—a feature observed in neurodegenerative diseases such as Parkinson’s and ALS.
