Background

Bassoon is a large (~420 kDa) presynaptic scaffold protein essential for the structural and functional organization of active zones in neuronal synapses. Localized at presynaptic terminals, Bassoon orchestrates the synaptic vesicle cycle by anchoring key components involved in neurotransmitter release and synaptic plasticity. Structurally, Bassoon contains two double-zinc finger motifs, three coiled-coil domains, and two polyglutamine (polyQ) stretches in its C-terminal region. These polyQ domains are of particular interest in neurodegenerative disease research, as abnormal expansions—similar to those observed in Huntington’s disease—may contribute to late-onset neurodegeneration through protein aggregation and synaptic dysfunction. In neurons, Bassoon is concentrated at sites directly opposite postsynaptic densities and co-localizes with both GABA (A) and glutamate (GluR1) receptors, highlighting its role in maintaining excitatory and inhibitory synaptic balance. Disruption of Bassoon expression or function has been linked to synaptic instability, altered neurotransmission, and increased vulnerability to neurodegenerative processes. Emerging evidence suggests that Bassoon may serve as a molecular hub in the early pathogenesis of disorders such as Alzheimer’s disease, Parkinson’s disease, and frontotemporal dementia, where synaptic loss is a key pathological hallmark. Its involvement in synaptic maintenance and plasticity positions Bassoon as a promising target for therapeutic strategies aimed at preserving neural connectivity in aging and disease.
