Background

In both prokaryotic and eukaryotic organisms, molecular chaperones play a vital role in preventing protein misfolding and aggregation during biogenesis and under stress conditions. Among these, the HSP60 family—also known as Cpn60 or GroEL in bacteria—is one of the most evolutionarily conserved and functionally essential. In Plasmodium falciparum, the causative agent of the most severe form of human malaria, HSP60 is a mitochondrial chaperone critical for parasite survival and adaptation to host-induced stress. PfHSP60 shares significant sequence homology with its bacterial and mammalian counterparts, particularly in its ATPase domain and oligomeric structure. Like other HSP60s, it forms homo-oligomeric complexes (typically 7 or 14 subunits) and exhibits ATP-dependent folding activity. These complexes are essential for the proper folding and assembly of mitochondrial proteins, especially under febrile conditions encountered during malaria infection. Studies have shown that PfHSP60, in cooperation with its co-chaperonin PfHSP10, can functionally complement the bacterial GroEL/GroES system, underscoring its evolutionary conservation and biological importance. Beyond its role in proteostasis, PfHSP60 may contribute to immune evasion and inflammation, and has been implicated in host-pathogen interactions. Given its essential function and stress-inducible nature, PfHSP60 represents a promising target for antimalarial drug development and a model for understanding mitochondrial chaperone systems in parasitic protozoa.
