Background

Phosphothreonine is a phosphorylated derivative of threonine, catalyzed by serine/threonine kinases. It serves as a regulatory switch in numerous signaling pathways that govern neuronal survival, synaptic function, and stress responses. In the brain, phosphothreonine modifications influence the activity of key proteins such as tau, MAPKs, and transcription factors. Dysregulated threonine phosphorylation is associated with protein misfolding, impaired axonal transport, and neuroinflammation—hallmarks of neurodegenerative diseases. Notably, abnormal phosphorylation of tau at threonine residues contributes to its aggregation and toxicity in Alzheimer’s disease.
