ASPN protein acts as a negative regulator in periodontal ligament (PDL) differentiation and mineralization, preventing ossification and maintaining homeostasis of the tooth support system. It inhibits BMP2-induced cell differentiation by blocking the binding of BMP2 to BMPR1B, thereby preventing BMP-dependent activation of SMAD proteins.
