Background

LGI1 (Leucine-Rich Glioma Inactivated 1) is a secreted synaptic protein that modulates neuronal excitability and synaptic transmission. It interacts with ADAM22 and ADAM23 to stabilize AMPA receptor function and maintain synaptic integrity. Mutations in LGI1 are linked to autosomal dominant lateral temporal lobe epilepsy (ADTLE) and have been implicated in autoimmune encephalitis. In neurodegenerative disease research, LGI1 is gaining attention for its role in synaptic maintenance, neuroinflammation, and neuronal survival. Dysregulation of LGI1 may contribute to cognitive decline and seizure susceptibility. Its dual role in epilepsy and neurodegeneration highlights LGI1 as a valuable biomarker and therapeutic target in synaptic pathologies.
