Background

LRRK2 (Leucine-Rich Repeat Kinase 2) is a large multidomain protein with kinase and GTPase activity, implicated in the pathogenesis of Parkinson’s disease (PD) and other neurodegenerative disorders. Mutations in LRRK2, such as G2019S, are among the most common genetic causes of familial and sporadic PD. LRRK2 regulates vesicle trafficking, autophagy, cytoskeletal dynamics, and immune responses. Its dysregulation leads to mitochondrial dysfunction, impaired lysosomal degradation, and neuroinflammation. LRRK2 is expressed in neurons and glial cells, and its activity is modulated by cellular stress and inflammatory signals. As a central node in neurodegenerative signaling networks, LRRK2 is a high-priority target for disease-modifying therapies in Parkinson’s and related disorders.
