Background

Notch1 is a transmembrane receptor involved in cell fate determination, synaptic plasticity, and neurogenesis. Following ligand binding, Notch1 undergoes sequential proteolytic cleavages, culminating in the release of the Notch intracellular domain (NICD), which translocates to the nucleus to regulate gene expression. In the nervous system, Notch1 signaling influences neural stem cell maintenance, differentiation, and response to injury. Aberrant Notch1 activation has been implicated in neurodegenerative diseases, where it may contribute to gliosis, inflammation, and impaired neurogenesis. Modulating Notch1 signaling could offer therapeutic strategies for promoting neural repair and mitigating neurodegenerative progression.
