Background

SNAT1, encoded by the SLC38A1 gene, is a sodium-coupled neutral amino acid transporter primarily responsible for glutamine uptake in neurons. As a key component of System A transporters, SNAT1 supports neurotransmitter cycling, energy metabolism, and nitrogen balance in the central nervous system. Highly expressed in specific neuronal populations—but notably absent in astrocytes—SNAT1 facilitates the intracellular accumulation of glutamine, a precursor for glutamate and GABA synthesis. This function is essential for maintaining excitatory and inhibitory neurotransmission, particularly in metabolically active brain regions. Disruption of SNAT1 expression or function has been linked to altered glutamate homeostasis, excitotoxicity, and oxidative stress—hallmarks of neurodegenerative diseases such as Alzheimer’s, Parkinson’s, and Huntington’s disease. Its role in retinal glutamate generation also suggests broader implications in neuro-ophthalmic disorders.
