Background

Matrix metalloproteinase 9 (MMP9) is an extracellular endopeptidase involved in the degradation of the extracellular matrix (ECM) . In the central nervous system, MMP9 regulates synaptic remodeling, neuroinflammation, and blood-brain barrier permeability. Elevated MMP9 activity has been observed in neurodegenerative diseases such as Alzheimer’s disease, multiple sclerosis, and amyotrophic lateral sclerosis (ALS) . MMP9 contributes to neuronal injury by promoting ECM breakdown, facilitating leukocyte infiltration, and activating pro-inflammatory cytokines. MMP9 is synthesized as a latent proenzyme and requires proteolytic cleavage for activation. Its expression is tightly regulated at the transcriptional level and by endogenous inhibitors such as TIMPs (tissue inhibitors of metalloproteinases) . Targeting MMP9 activity offers therapeutic potential for preserving blood-brain barrier integrity, reducing neuroinflammation, and limiting tissue damage in neurodegenerative conditions.
