Background

Nav1.7 is a voltage-gated sodium channel encoded by the SCN9A gene and is predominantly expressed in peripheral sensory neurons. It plays a pivotal role in pain perception by amplifying subthreshold stimuli and facilitating action potential initiation. Loss-of-function mutations in Nav1.7 result in congenital insensitivity to pain, while gain-of-function mutations are associated with chronic pain syndromes. Importantly, Nav1.7 is not expressed in cardiac or central neurons, making it an attractive target for pain therapeutics with minimal off-target effects. Although primarily studied in nociception, emerging research suggests that peripheral nerve dysfunction and chronic pain may contribute to neurodegenerative disease burden. Nav1.7 inhibitors are under investigation as next-generation analgesics with potential applications in neurodegenerative conditions involving neuropathic pain.
