Background

Cav1.3, encoded by the CACNA1D gene, is a voltage-gated L-type calcium channel subunit predominantly expressed in neurons and neuroendocrine cells. It plays a vital role in regulating calcium influx, which is essential for neuronal excitability, synaptic plasticity, gene expression, and neurosecretion. In the central nervous system, Cav1.3 contributes to pacemaker activity in dopaminergic neurons and modulates long-term potentiation—key processes underlying learning and memory. Dysregulation of Cav1.3-mediated calcium signaling has been implicated in several neurodegenerative and neuropsychiatric disorders, including Parkinson’s disease, Alzheimer’s disease, and autism spectrum disorders.
