Background

FKBP52 (FK506-binding protein 4) is a co-chaperone that interacts with Hsp90 and steroid hormone receptors, influencing protein trafficking, microtubule dynamics, and neuronal signaling. In the nervous system, FKBP52 plays a protective role by regulating tau protein conformation and preventing its pathological aggregation. Reduced FKBP52 levels have been linked to increased tau pathology in Alzheimer’s disease, suggesting a neuroprotective function. FKBP52 also modulates axonal transport and synaptic stability, processes that are disrupted in several neurodegenerative conditions. As a regulator of both protein homeostasis and cytoskeletal integrity, FKBP52 represents a promising therapeutic target for halting or reversing neurodegenerative processes.
