Human Runt-related transcription factor 3 (RUNX3) ELISA Kit

RUNX3 is a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex that binds to the core DNA sequence 5'-PYGPYGGT-3' found in a number of enhancers and promoters, and can either activate or suppress transcription. It also interacts with other transcription factors. It functions as a tumor suppressor, and the gene is frequently deleted or transcriptionally silenced in cancer. Multiple transcript variants encoding different isoforms have been found for this gene. Runx3 mouse gastric mucosa exhibits hyperplasia due to stimulated proliferation and suppressed apoptosis in epithelial cells, and the cells are resistant to TGF-beta stimulation.

Human Runt-related transcription factor 3 (RUNX3) ELISA Kit has high sensitivity and excellent specificity for detection of Human RUNX3. No significant cross-reactivity or interference between Human RUNX3 and analogues was observed.

This Human Runt-related transcription factor 3 (RUNX3) ELISA Kit employs a two-site sandwich ELISA to quantitate RUNX3 in samples. An antibody specific for RUNX3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRUNX3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RUNX3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RUNX3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
