Human Protein Wnt-9b (WNT9B) ELISA Kit

WNT9B is a member of the WNT gene family. Study of its expression in the teratocarcinoma cell line NT2 suggests that it may be implicated in the early process of neuronal differentiation of NT2 cells induced by retinoic acid. This gene is clustered with WNT3, another family member, in the chromosome 17q21 region.WNT15 shows 53% amino acid identity to chicken Wnt14, 54% identity to human WNT14, and 65% identity to shark Wnt9. The deduced 359-amino acid protein shares significant identity with the 331-amino acid human WNT9B protein, and both proteins contain all 23 cysteines conserved in Wnt proteins. Northern blot analysis of adult mouse tissues detected highest expression in kidney and lower expression in liver, brain, male preputial gland, and female mammary gland. No expression was detected in other adult tissues examined.

Human Protein Wnt-9b (WNT9B) ELISA Kit has high sensitivity and excellent specificity for detection of Human WNT9B. No significant cross-reactivity or interference between Human WNT9B and analogues was observed.

This Human Protein Wnt-9b (WNT9B) ELISA Kit employs a two-site sandwich ELISA to quantitate WNT9B in samples. An antibody specific for WNT9B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyWNT9B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WNT9B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT9B bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Do not mix components from different kit lots or use reagents beyond the kit expiration date.• Allow all reagents to warm to room temperature for at least 30 minutes before opening.• Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.• It is recommended that all samples and standards be assayed in duplicate or triplicate.
