SLC40A1 (Human) ELISA Kit
Ferroportin-1 (FPN1, IREG1, SLC40A1) is a transmembrane protein that belongs to the ferroportin family of proteins. It is the only known iron exporter in mammals. After absorption of dietary iron in the small intestine, SLC40A1 exports the iron out of the intestinal cells into the bloodstream. It is expressed in cells such as macrophages, hepatocytes, duodenal enterocytes, placental syncytiotrophoblasts, and adipocytes. However, it is absent in erythroid cells. Expression of SLC40A1 is regulated by Hepcidin, a liver-derived hormone that causes internalization and degradation of SLC40A1 and reduces the efflux of iron into the bloodstream. Both Hepcidin and SLC40A1 play a critical role in the pathogenesis of iron metabolism diseases, iron overload, and iron-limited anemia. Mutations in SLC40A1 result in an autosomal dominant disorder called type IV hemochromatosis or Ferroportin Disease. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of SLC40A1 in the sample or standard.
