PER1 (Human) ELISA Kit
Period circadian protein homolog 1 (PER1) is a master regulator of the circadian clock that belongs to the period family of proteins. It is expressed in almost all the cells of peripheral tissues, the most notable expression being in the suprachiasmatic nucleus (SCN), a primary circadian pacemaker in the brain. PER1 is involved in a transcription-translation feedback loop wherein it is activated by CLOCK-BMAL1 heterodimer inside the nucleus. PER1 then heterodimerizes with CRYs in the cytoplasm and reenters the nucleus to inhibit CLOCK-BMAL1 transcriptional activity. Besides playing a critical role in the entrainment of the circadian clock to the light-dark cycles, PER1 also regulates the cell cycle and response to DNA damage. Aberrant expression or altered rhythms of PER1 were associated with multiple cancers such as prostate, colon, breast, head and neck, and endometrial cancer. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of PER1 in the sample or standard.
