Prosaposin (Human) ELISA Kit
Prosaposin (PSAP) is a 66 kDa glycoprotein that exists as a common lysosomal precursor for Saposins. It is proteolytically processed in the lysosomes to yield Saposins A-D. Together with four saposins, PSAP activates lysosomal hydrolases that mediate hydrolysis of glycosphingolipids and sphingolipids. In addition to being an integral membrane protein, PSAP also exists as a secreted protein in biological fluids such as cerebrospinal fluid, milk, semen, serum, bile, and pancreatic juice wherein they perform different physiological roles. Besides activating lysosomal hydrolases, PSAP is involved in the development of the reproductive and nervous system. Deficiency of PSAP is associated with lysosomal and lipid storage diseases. On the other hand, increased expression of PSAP has been observed in prostate, breast, and gallbladder cancer. Due to its diverse roles in biological processes, PSAP could be a promising target for the treatment of human diseases. BioVision’s PSAP (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of PSAP in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of PSAP in the sample or standard.
