Background

IL-17C (Interleukin 17C) is a pro-inflammatory cytokine that plays a crucial role in epithelial innate immunity, especially against intestinal bacterial pathogens, acting in an autocrine manner. It stimulates the production of antibacterial peptides and pro-inflammatory molecules for host defense via activating the NFKB and MAPK signaling pathways. It acts synergistically with IL22, TNF and IL1B to induce antibacterial peptides. It may exert a protective effect by maintaining epithelial homeostasis after inflammatory challenges, such as intestinal inflammation in a dextran sulfate sodium-induced colitis model. It can also promote an inflammatory phenotype, e.g., in psoriatic skin. Enhanced IL17C/IL17RE signaling may increase susceptibility to autoimmune diseases like autoimmune encephalitis.

Assay Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IL-17C. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse IL-17C and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse IL-17C, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 7 nm. The OD value is proportional to the concentration of Mouse IL-17C. You can calculate the concentration of Mouse IL-17C in the samples by comparing the OD of the samples to the standard curve.
