Background

Macrophage Inflammatory Protein 5 (MIP-5), also designated as C-C motif chemokine ligand 15 (CCL15), is a member of the CC chemokine family, initially discovered by analyzing the upstream region of the CCL14 gene. Located on human chromosome 17, it is arranged in tandem with CCL14 and belongs to the NC6 beta-chemokines, featuring an extended N-terminal domain (16-20 residues) encoded by a distinct exon upstream of the first two conserved cysteines, shared with CCL6, CCL9, and CCL23. The precursor CCL15 (1-92) undergoes proteolytic processing by cathepsin G, elastase, and MT6-MMP/MMP25 to generate more active truncated forms, such as CCL15 (22-92), (24-92), (27-92), and (29-92) . As a chemotactic factor, MIP-5/CCL15 primarily attracts T-cells and monocytes (not neutrophils, eosinophils, or B-cells) via binding to CC chemokine receptor 1 (CCR1), with additional binding to CCR3, and its truncated forms exhibit stronger chemoattractant activity. Clinically, it is closely associated with various diseases: elevated in renal cell carcinoma bone metastasis, pulmonary sarcoidosis bronchoalveolar lavage, severe asthma serum (reduced by omalizumab), and hepatocellular carcinoma (HCC) serum, where it promotes tumor progression via the CCL15-CCR1 axis. It is also linked to colorectal cancer and esophageal squamous cell carcinoma, highlighting its potential as a diagnostic and therapeutic target.

Assay Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human MIP-5. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human MIP-5 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human MIP-5, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 3 nm. The OD value is proportional to the concentration of Human MIP-5. You can calculate the concentration of Human MIP-5 in the samples by comparing the OD of the samples to the standard curve.
