BH4 (Tetrahydrobiopterin) ELISA Kit

Tetrahydrobiopterin (BH4) is an important coenzyme involved in various biochemical processes, including serotonin, dopamine, norepinephrine, adrenaline, and melatonin. It has the function of antioxidant and scavenging active nitrogen oxides in human body, and plays a key role in diabetes, pulmonary hypertension, pathological myocardial remodeling and other diseases. In addition, BH4 has been found to play a dual role in promoting and inhibiting the occurrence and development of malignant tumors. Tetrahydrobiopterin deficiency is a rare autosomal inherited disease, which belongs to hyperphenylalaninemia (HPA) along with phenylketonuria. Its characteristic is insufficient synthesis of monoamine neurotransmitters dopamine and serotonin due to BH4 biosynthesis or circulatory disorders. This disease can be better controlled by delaying diagnosis. In terms of treatment, the application of BH4 in the treatment of phenylketonuria has shown its significant ability to reduce blood Phe levels and increase dietary tolerance, which is crucial for partial or even complete relaxation of strict dietary control. Meanwhile, studies have shown that BH4 induces resistance to proteasome inhibitors and tumor progression in multiple myeloma, and its key role in radiosensitivity regulation has also been confirmed.

This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal BH4. During the reaction, Universal BH4 in the sample or standard competes with a fixed amount of Universal BH4 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal BH4. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal BH4 in tested samples can be calculated by comparing the OD of the samples to the standard curve.
