Background
Interleukin-5 (IL-5) is a homodimeric cytokine, where two identical 126-amino-acid monomers are linked by a disulfide bond. It is primarily secreted by activated Th2 cells, with additional sources including eosinophils, mast cells, iNKT cells, and NK cells.IL-5 is the principal regulator of eosinophils, driving their differentiation, survival, activation, and chemotaxis. It also activates basophils. Beyond granulocytes, IL-5 acts on B cells to promote immunoglobulin production, growth, and differentiation, linking it to humoral immunity. Its biological effects are mediated by a cell surface receptor complex composed of a unique IL-5Rα chain and the common beta chain (βc/CD131) . Receptor engagement activates JAK2 and other kinases, triggering downstream JAK-STAT5 and RAS-MAPK signaling pathways. Bioavailability is naturally regulated by soluble IL-5Rα, which acts as a decoy receptor. Thus, IL-5 is a central cytokine in type 2 immunity, allergic inflammation, and antibody responses, making it a key therapeutic target for eosinophil-driven diseases.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IL-5. Samples (or Standards &Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse IL-5 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse IL-5, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Mouse IL-5. You can calculate the concentration of Mouse IL-5 in the samples by comparing the OD of the samples to the standard curve.
