Background
Interleukin-17A (IL-17A) is a 15-20 kDa glycosylated, pro-inflammatory cytokine and a central member of the IL-17 family (A-F) . It is primarily secreted by T helper 17 (Th17) cells but also by innate immune cells like γδ T cells, iNKT cells, and group 3 innate lymphoid cells (ILC3s) . IL-17A signals through a heterodimeric receptor complex of IL-17RA and IL-17RC, activating NF-κB and MAPK pathways via the adaptor TRAF3IP2. This induces the expression of chemokines (e.g., CXCL1, CXCL5), antimicrobial peptides (e.g., defensins), and matrix metalloproteinases. Its primary function is to bridge adaptive and innate immunity by robustly recruiting and activating neutrophils to clear extracellular bacterial and fungal infections. It also maintains epithelial barrier integrity and promotes mucosal host defense. Furthermore, IL-17A regulates B cell responses in germinal centers and participates in antiviral immunity. IL-17A can form homodimers or heterodimers with the closely related IL-17F (sharing 50% homology) . While both homodimers and the heterodimer are bioactive, IL-17A is the most potent driver of neutrophil migration. Beyond host defense, IL-17A has a dual role in pathology. It is a key driver of chronic inflammation and autoimmunity (e.g., rheumatoid arthritis, psoriasis) and can promote tumorigenesis. Conversely, it can exhibit protective functions in colitis and exert anti-tumor effects in certain contexts, making it a complex and critical therapeutic target.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IL-17A. Samples (or Standards &Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse IL-17A and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse IL-17A, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Mouse IL-17A. You can calculate the concentration of Mouse IL-17A in the samples by comparing the OD of the samples to the standard curve.
