Background
Interleukin-1 (IL-1) comprises two proteins, IL-1α and IL-1β, encoded by separate genes but functioning similarly. Both are synthesized as 31 kDa precursors lacking a signal peptide and processed to ~17 kDa mature forms. IL-1α is often active as a precursor and retained intracellularly or membrane-bound, whereas IL-1β requires caspase-1 cleavage for activation and release.Produced by monocytes, macrophages, fibroblasts, endothelial cells, and neural cells, IL-1 signals mainly through IL-1 receptor type I (IL-1RI) . IL-1 receptor type II (IL-1RII) acts as a decoy, inhibiting signaling by sequestering IL-1.A key mediator of innate immunity and inflammation, IL-1 induces fever, immune cell activation, cytokine release, and acute-phase responses. It also influences hematopoiesis, bone metabolism, and contributes to autoimmune, vascular, and neurodegenerative diseases. Its activity is regulated transcriptionally, post-translationally, via receptor balance, and by antagonists like IL-1Ra.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IL-1α. Samples (or Standards & Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse IL-1α and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse IL-1α, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Mouse IL-1α. You can calculate the concentration of Mouse IL-1α in the samples by comparing the OD of the samples to the standard curve.
