Background
Vascular endothelial growth factor (VEGF-A/VEGF), originally termed vascular permeability factor (VPF), is a key regulator of angiogenesis and vasculogenesis during development and in adulthood. It belongs to the PDGF family, sharing the characteristic cystine knot motif and functioning as a disulfide-linked homodimer.VEGF is widely expressed in various tissues and cell types—including muscle, hepatocytes, osteoblasts, neutrophils, macrophages, and endothelial cells—and its production is induced by hypoxia and cytokines such as IL-1, IL-6, and TNF-α.During embryogenesis, VEGF drives endothelial cell proliferation, migration, and survival, thereby regulating vessel density and size. Postnatally, it supports endothelial integrity, wound healing, and female reproductive cycling. Pathologically, VEGF enhances vascular permeability and promotes tumor angiogenesis and metastasis. It also contributes to autoimmune conditions like rheumatoid arthritis and multiple sclerosis, making it a target for anti-angiogenic therapies.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse VEGF-A. Samples (or Standards &Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse VEGF-A and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse VEGF-A, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Mouse VEGF-A. You can calculate the concentration of Mouse VEGF-A in the samples by comparing the OD of the samples to the standard curve.
