Background
Interleukin-1β (IL-1β) is a potent master pro-inflammatory cytokine. Synthesized as an inactive precursor, its maturation is tightly regulated, primarily by caspase-1 via inflammasome activation or directly by microbial proteases. Active IL-1β is released through non-classical pathways like Gasdermin-D pores. It drives systemic inflammation, fever, immune activation (including Th17 differentiation), and angiogenesis, playing a central role in host defense. However, its activity is strictly controlled by natural inhibitors like IL-1 receptor antagonist (IL-1ra) and decoy receptors. Dysregulated or excessive IL-1β production is a key pathogenic driver in numerous diseases, including rheumatoid arthritis, atherosclerosis, and diabetes, making it a critical therapeutic target.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IL-1β. Samples (or Standards & Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse IL-1β and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse IL-1β, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±10 nm. The OD value is proportional to the concentration of Mouse IL-1β. You can calculate the concentration of Mouse IL-1β in the samples by comparing the OD of the samples to the standard curve.
