Background
IL-12 p70 is the biologically active heterodimer of interleukin-12 (IL-12), consisting of disulfide-linked IL-12A (p35) and IL-12B (p40) subunits with a molecular weight of approximately 70 kDa. It is primarily produced by professional antigen-presenting cells (APCs) following antigen stimulation, which distinguishes it from inactive p40 monomers or homodimers. Functionally, IL-12 p70 induces interferon-gamma (IFN-γ) production, promotes the differentiation of naive T cells into T-helper 1 (Th1) cells, and enhances the activity of natural killer (NK) cells and cytotoxic T lymphocytes (CTLs) . It exerts its effects by binding to the IL12Rβ1/β2 heterodimeric receptor, which activates JAK-STAT4 signaling to regulate cytokine-responsive gene expression.IL-12 p70 shares the p40 subunit with IL-23 and the p35 subunit with IL-35. As a key link between innate and adaptive immunity, it plays a vital role in research on infectious diseases, autoimmune disorders, and cancer immunotherapy.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-12. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-12 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-12, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 19 nm. The OD value is proportional to the concentration of Human IL-12. You can calculate the concentration of Human IL-12 in the samples by comparing the OD of the samples to the standard curve.
