Background
Interleukin-23 (IL-23) is a heterodimeric cytokine pivotal in bridging innate and adaptive immunity. It is composed of two disulfide-linked subunits: a unique p19 subunit (IL-23A) and a p40 subunit (IL-12B) shared with IL-12. While p19 is expressed by various activated immune and endothelial cells, the concurrent expression of p40—primarily by activated macrophages and dendritic cells—is the critical limiting step for functional IL-23 production.IL-23 signals through a heterodimeric receptor complex consisting of IL-12Rβ1 and the IL-23-specific IL-23R. This interaction activates the JAK2/STAT signaling cascade, leading to phosphorylation of STAT1, STAT3, STAT4, and STAT5.Functionally, IL-23 has distinct roles from IL-12. Its action is largely restricted to memory T cells, driving their proliferation and, crucially, their production of the potent pro-inflammatory cytokine IL-17. This IL-23/IL-17 axis is a central mediator of inflammatory and autoimmune pathogenesis. Additionally, IL-23 can enhance immunogenic antigen presentation by CD8+ dendritic cells. Thus, IL-23 is a key regulator of inflammatory responses, critically involved in host defense, autoimmune diseases, and tumor immunology.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-23. Samples (or Standards & Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-23 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-23, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±6 nm. The OD value is proportional to the concentration of Human IL-23. You can calculate the concentration of Human IL-23 in the samples by comparing the OD of the samples to the standard curve.
