Background
Interleukin-22 (IL-22), initially identified as an IL-9-inducible gene (IL-TIF), is a member of the IL-10 cytokine family. The human protein is produced as a 179-amino-acid precursor, cleaved to a 147-aa active form, sharing ~22% and ~79% sequence identity with human IL-10 and mouse IL-22, respectively. Its gene is located on chromosome 12q15. IL-22 signals through a heterodimeric cell surface receptor composed of IL-22R1 (CRF2-9) and IL-10R2 (CRF2-4), activating STAT1 and STAT3 pathways. Its activity is further modulated by a soluble inhibitor, IL-22 binding protein (IL-22BP) . Primarily produced by immune cells like activated T cells and Th17 lymphocytes, IL-22 acts predominantly on non-immune epithelial and stromal cells. It plays a critical role in mucosal host defense, tissue repair, and the induction of acute-phase proteins. However, it also contributes to the pathogenesis of chronic inflammatory and autoimmune diseases, highlighting its dual role in regulating tissue homeostasis and inflammation.

Assay Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-22. Samples (or Standards & Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-22 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-22, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±5 nm. The OD value is proportional to the concentration of Human IL-22. You can calculate the concentration of Human IL-22 in the samples by comparing the OD of the samples to the standard curve.
