CellaQuantTM Human IL-2 (Interleukin 2) ELISA Kit

Interleukin-2 (IL-2), or T-cell growth factor (TCGF), is a 15–18 kDa glycosylated cytokine in the common gamma chain (γc) family, with a short half-life under 30 minutes. It is produced by activated CD4+and CD8+T cells, γδT cells, microglia, and hematopoietic stem cells.The IL-2 receptor consists of CD25 (IL-2Rα), IL-2Rβ, and γc subunits. IL-2 binding to CD25 recruits IL-2Rβ and γc to form a signaling complex. IL-2Rβ is also used by IL-15, and γc is shared by multiple cytokines including IL-4, IL-7, and IL-15.IL-2 drives T cell activation and expansion in vitro. In vivo, it is essential for regulatory T cell (Treg) development and function, preventing autoimmunity, but can also promote inflammatory responses via Th2 cytokines and trafficking gene regulation. In CD8+T cells, IL-2 enhances effector differentiation and memory formation.

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-2. Samples (or Standards & Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-2 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-2, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±3 nm. The OD value is proportional to the concentration of Human IL-2. You can calculate the concentration of Human IL-2 in the samples by comparing the OD of the samples to the standard curve.

This ELISA kit applies to the in vitro quantitative determination of Human IL-2 concentrations in Cell culture supernatant, Cell lysates or other biological fluids.
